An EPR study of the production of superoxide radicals by neutrophil NADPH oxidase.

چکیده
Superoxide production by stimulated neutrophils and by preparations of NADPH oxidase has consistently been measured by an indirect method utilising the reduction of cytochrome c by superoxide radicals [4]. This reduction is inhibited by superoxide dismutase. EPR spectroscopy using the technique of spin trapping has been used to identify superoxide radicals formed by photo-irradiated spinach chloroplasts [5] and human neutrophils following stimulation [6]. This technique appears to be the least unambiguous method of detecting superoxide production by biological systems in their normal physiological state. Here, we provide the first direct demonstration of superoxide production by guinea-pig neutrophil NADPH oxidase. The results indicate that as seen by spin trapping, the superoxide radical is the only free radical produced by the enzyme in the presence of NADPH. 2. MATERIALS AND METHODS

The generation of hydroxyl radicals following superoxide production by neutrophil NADPH oxidase.
Stimulation of neutrophils with membrane perturbing agents causes an increase in oxygen consumption concomitant with the production of superoxide and hydroxyl radicals and hydrogen peroxide (HzOz). A plasma membrane-bound NADPH oxidase which is inactive in resting neutrophils has been implicated in the production of these radicals [ 1,2]. Purified NADPH oxidase has recently been shown to produc...
متن کاملExpression of NADPH oxidase in rabbit corneal epithelial and stromal cells in culture.
PURPOSE Reactive oxygen- and nitrogen-containing molecules produced in high concentrations are mediators of tissue damage caused by inflammation. The free radical molecules superoxide (O2-*) and nitric oxide (NO*), when produced at low concentrations, may function as second messengers or regulators of signal transduction. The purpose of these studies was to determine whether corneal epithelial ...
متن کاملAldosterone Induces Oxidative Stress Via NADPH Oxidase and Downregulates the Endothelial NO Synthesase in Human Endothelial Cells
Aldosterone is traditionally viewed as a hormone regulating electrolyte and blood pressure homeostasis. Recent studies suggest that Aldo can cause microvascular damage, oxidative stress and endothelial dysfunction. However, its exact cellular mechanisms remain obscure. This study was undertaken to examine the effect of Aldo on superoxide production in human umbilical artery endothelial cel...
متن کاملNADPH oxidase-derived reactive oxygen species in skeletal muscle modulates the exercise pressor reflex.
Muscle metabolic by-products during exercise, such as K+, lactic acid, ATP, H+, and phosphate, are well established to be involved in the reflex cardiovascular response to static muscle contraction. However, the role of muscle reactive oxygen species (ROS), a metabolic by-product during muscle contraction, in the exercise pressor reflex (EPR) has not been investigated in detail. In the present ...

OBJECTIVES The production of reactive oxygen species (e.g., superoxide) by endothelial cells is relevant to tissue injury during ischemia-reperfusion, and may also play a role in intracellular signaling pathways. However, the molecular identities of the enzymes responsible for endothelial superoxide production are poorly defined, although xanthine oxidase, NADH/NADPH oxidoreductases and nitric ...
